Sylvain Baillet is a physicist with research interests in methodological developments for functional and structural brain imaging and their application to clinical and cognitive Neuroscience. Although his primary expertise is with MEG (magnetoencephalography) imaging, he also contributed new techniques for MRI image segmentation and inter-subject registration. He has also initiated new image analysis techniques for the early prediction of infarct growth in stroke patients, using diffusion-weighted MR sequences. His current research projects with MEG include real-time therapeutic imaging, the evaluation of resting-state brain activity and connectivity in the healthy and diseased brain, and the development of original data mining and visualization techniques for time-resolved brain image series. He has also great interest in disseminating his research through academic software: he is a founding developer of the Brainstorm open-source software project (http://neuroimage.usc.edu/brainstorm) for MEG and EEG imaging with MRI integration.

Profile

After completing a post-doctoral research fellowship training at the Biological Psychiatry Branch of the National Institutes of Health (Bethesda, MD, USA) with Robert M. Post, MD, Serge Beaulieu, MD, PhD, FRCPC, joined the Douglas Institute in 1997 and assumed the post of director of the Mood Disorders Programs. At McGill University, he is an associate professor in the Department of Psychiatry and an associate member of the Department of Neurology and Neurosurgery.

A psychiatrist and researcher-clinician, Serge Beaulieu is interested primarily in bipolar disorders (manic depressive illness) and major depression. He studies neurobiological responses to antidepressant and mood stabilizer treatment and potential new treatments for these disorders.

Serge Beaulieu has received diverse peer-reviewed research funding from the National Alliance for Research on Schizophrenia and Affective Disorders (NARSAD, USA), the Stanley Foundation (USA), the Fonds de la recherche en santé du Québec (FRSQ), the Canadian Fund for Innovation (CFI) and the Canadian Institutes of Health Research (CIHR).

He is a member of the board of governors of the Canadian Network on Mood and Anxiety Treatment (CANMAT), a member of the Board of Councilors of the International Society of Bipolar Disorders (ISBD) and one of the investigators and founding members of the Canadian Bipolar Consortium.

Barry Bedell
Dr. Barry Bedell joined the faculty at the Montreal Neurological Institute and Hospital in July 2006 as an Assistant Professor in the Department of Neurology and Neurosurgery at McGill University. He is the first recruit to accept a position as part of the Montreal Consortium for Brain Imaging Research (MCBIR) initiative, which is a partnership between the MNI, McGill, the Douglas Hospital Research Centre and the Lady Davis Institute of the Jewish General Hospital.

A native of New Jersey, Dr. Bedell holds a B.S. (Chemistry) from Lehigh University (1992) and a Ph.D. in Medical Physics (1996) from the University of Texas Health Science Center at Houston. He obtained his M.D.C.M. from McGill University in 2001. While pursing his medical degree and completing his residency in pathology, Dr. Bedell conducted research with Dr. Alan Evans and colleagues in the McConnell Brain Imaging Centre.

Dr. Bedell will lead the effort to develop the animal imaging research program which will be housed in the new addition to the BIC in The Neuro's North Wing. His current research program is the development of an Integrated Technology Platform (ITP), which integrates MRI/PET imaging, histopathological, and molecular studies in animal models of Alzheimer's disease, multiple sclerosis (MS), and brain tumours. As part of this multi-disciplinary research, Dr. Bedell has recently established a state-of-the-art experimental neuropathology facility which focuses on high-throughput production of quantitative data from tissue sections and seamless integrates with the animal imaging activities.

David Benrimoh is a neuropsychiatrist who completed his psychiatry residency at McGill. He received an MSc. in Neuroscience from UCL, working with Karl Friston on computational models of auditory hallucinations. He received a second MSc. in Psychiatry from McGill, working with Simon Ducharme and Bratislav Misic on transdiagnostic imaging in psychosis. He is the founder and chief science officer at Aifred Health, a digital mental health company using artificial intelligence to create decision support tools for mental health clinicians; Aifred won the $1M USD second place prize in the global IBM-sponsored AI XPRIZE. He completed his fellowship in Neuropsychiatry at Stanford University, where he conducted research on the mechanistic underpinnings of rapid acting rTMS. His research lab, the McGill Lab for Computational Psychiatry and Translation (McPsyt), focuses on computational psychiatry approaches to understanding and predicting psychosis onset and using this knowledge to develop novel treatment paradigms using rTMS.

Leukodystrophies are a group of over 20 genetically-determined disorders characterized by abnormal white matter on brain imaging. Typically, they are neurodegenerative diseases starting in previously healthy children, leading to progressive disabilities and death over months to years. They are classified into two categories, according to whether the primary problem is maintaining myelin homeostasis (non-hypomyelinating) or myelin deposition during development (hypomyelinating). I have built a comprehensive research program aimed at characterizing leukodystrophies clinically, radiologically, genetically and pathophysiologically. My research program is expanding to include perspectives from the study of epidemiology, natural history, and quality-of-life impacts, with the ultimate objective of improving care for children who face the serious challenges associated with this disease group. My research program focuses on hypomyelinating leukodystrophies, and more specifically on 4H or POLR3-related leukodystrophy. 4H is a hypomyelinating leukodystrophy characterized by neurological (i.e. cerebellar and pyramidal features, cognitive regression) and non-neurological (i.e. myopia, dental and endocrine abnormalities) characteristics. It is caused by recessive mutations in POLR3A, POLR3B and POLR1C, encoding subunits of RNA-polymerase III, which is the enzyme responsible for the transcription of small non-coding RNA important for primordial cellular processes. Using 4H leukodystrophy as the primary disease of interest, my research translational program has the following objectives: (1) Clinical and radiological characterization of known and novel disorders, (2) Identification and validation of novel causal genes, (3) Understanding the impacts of the disease on patients and family members over time, and the factors involved in alleviating or worsening these, (4) Understand the cellular mechanisms involved in hypomyelination, and more specifically, the contribution of transcription and translation on hypomyelination (5) Using an in vitro model of hypomyelination, intervene to improve myelination.

The overall aim of this program is to understand the clinical, radiological, genetic, molecular and pathophysiological bases of hypomyelinating leukodystrophies to develop innovative treatments and rapidly translate them into clinical practice for these devastating neurodevelopmental and neurodegenerative diseases.

We develop cutting-edge computational approaches to key questions in genomics and evolution. We design algorithmic, machine learning, and statistical approaches to solve real-world questions from biology, and work in tight collaboration with biologists, biochemists, and geneticists to put our tools to use to make biological discoveries. We are particularly interested in: (i) How do genomes evolve, and what does that tell us about their functions? (ii) How is gene expression regulated, and what role does 3D chromosome organization play? Of particular interest in the context of our work are normal and cancer brain cells. We are studying how chromatin conformation evolves as cells differentiate from embryonic stem cells to neural progenitor cells to neuron, and how expression changes accompany chromatin structure changes. In brain cancer, we are working with Nada Jabado’s lab to map the 3D organization of chromatin in various types of paediatric brain tumours, identify markers of different types of cancer, and link these changes to other genetic and epigenetic changes, with the goal of identifying putative therapeutical targets.

Magnetic resonance imaging (MRI) is one of the tools of choice for the analysis of the brain in normally functioning and diseased states. Different MRI acquisition protocols provide excellent contrast and resolution and can be analyzed using automated techniques. The goal of Dr. Chakravarty’s work is to develop and use computational neuroanatomical techniques to understand how the structure of the brain is altered in psychiatric disorders. These techniques include the automated identification of structures and computational metrics that quantify their shape.  He is currently working on adding information from diffusion tensor imaging in order to describe inter-structural connectivity.  Many of these techniques can be used to elucidate phenotypes that can help describe brain risk for the major psychosis or pathological ageing.  To date, Dr. Chakravarty and the Computational Brain Anatomy (CoBrA) Laboratory have been using these techniques to better understand the alterations in brain anatomy that underly the major psychoses, ageing, brain development, and Alzheimer's disease.

Dr. Jean-François Cloutier’s research is aimed at understanding the molecular mechanisms that control axonal pathfinding during development of the nervous systems. The formation of accurate connections between the peripheral and central nervous systems is essential for the functioning of the various sensory systems that allow us to detect odors, sounds, tastes, pain, and temperature. While the basic anatomical principles of wiring in these systems are well understood, little is known about the mechanisms involved in establishing these wiring patterns.

Dr.Cloutier uses a combination of in vivo mouse genetic models and molecular biological approaches to identify cues involved in guiding axons of primary sensory neurons to their targets and to characterize intracellular signals that function downstream of these axon guidance cues. Gaining insight into the basic mode of action of guidance molecules will be important in the future development of therapies for neurological disorders and injuries requiring regeneration of specific axonal connections.

Our laboratory focuses in degenerative and regenerative processes in the CNS with particular emphasis on aging and Alzheimer’s disease (AD) related studies. The group utilizes and develops transgenic animal models presenting features of the AD neuropathology. The research is of a multidisciplinary nature ranging from molecular approaches to whole animal experimentation and with a particular interest in cell and animal models of the Alzheimer’s disease and experimental therapeutics.
Professor Cuello has been trained at the Universities of Buenos Aires (UBA, Argentina), U. C. San Francisco (USA) and Cambridge (England). He held research and academic positions  in the Departments of Pharmacology at both Cambridge and Oxford. He is a past Chair of the McGill Department of Pharmacology and Therapeutics and presently holds the Charles E. Frosst Merck Chair within the same Department. He is also currently an Associate Member of the Departments of Anatomy and Cell Biology and Neurology and Neurosurgery at McGill University, adjunct Professor of the Scripps Institute (LaJolla) and Visiting Professor at Oxford University.

Our lab focuses on studying cell dynamics in various biological processes in many diseases (e.g., developmental disorder, pulmonary diseases, cancers). Decoding cell dynamics is essential for understanding the pathogenesis of diseases and finding novel therapeutics. The existence of enormous heterogeneity in those diseases makes it challenging to decipher the unknown. The advancing single-cell technologies that profile individual cell states provide unprecedented opportunities to tackle this problem, which could drive biological discoveries and medical innovations in various fields (such as developmental and cancer biology). However, the single-cell data presents numerous new challenges in developing computational models that bridge the biomedical data and potential discoveries. Our primary research is to develop machine learning approaches (particularly probabilistic graphical models) to jointly analyze, model, and visualize single-cell (and/or bulk) omics data (preferably longitudinal or spatial). Such computational models will be used to help us derive a deeper understanding of the cell dynamics in different biological systems, which will eventually benefit the public health with machine-learning driven new diagnostic and therapeutic strategies.

The main focus of my research is on the brain circuitry that controls how we cope with stressors and its role in drug addiction. I am currently interested in the medial prefrontal cortex (PFC), a brain region that reaches its full development during late adolescence/early adulthood.  This brain region controls several complexe tasks such as decision-making and planning and has been implicated in the compulsive behavior of drug addicts.  In part because it matures slowly, the PFC is particularly vulnerable to insults which can cause it to develop abnormally and lead to maladaptive behavior. The PFC is also extremely sensitive to stressors and plays an important role in mounting the appropriate coping responses. The neurotransmitters, dopamine and glutamate, have been implicated in PFC-mediated coping responses to stressors and actions of addictive drugs.  For our studies we use voltammetry and microsensors to monitor, in behaving rats, minute changes in dopamine and glutamate transmission on a second-to-second basis.

Many essential cellular functions including cell division, motility, protein synthesis, and intracellular transport are driven by motor proteins, a specialized set of enzymes that convert chemical energy into mechanical work. Two motor proteins, kinesin and cytoplasmic dynein, are responsible for the long-range transport of mRNA, proteins, organelles, and signaling molecules along the microtubule cytoskeleton. Active transport by kinesin and dynein is critical for the maintenance of biosynthetic, signaling, and degradative pathways in the cell. Long and highly-polarized cells like neurons are particularly sensitive, and accordingly mutations in kinesin or dynein cause neurodegenerative disease in mouse models and humans. Further, defects in intracellular transport have been linked to many neurodegenerative diseases including amyotrophic lateral sclerosis, Alzheimer’s disease, and Huntington’s disease.

In the complex cellular environment, kinesin and dynein are regulated by interactions with the cytoskeleton, other motor proteins, and binding partners. These interactions allow motors to perform complicated functions such as cell division and the targeted trafficking of intracellular cargoes. The Hendricks lab is focused on understanding how motor proteins function collectively, and how interactions among motor proteins and with the complex cellular environment modulate their behavior. We employ in vitro experiments that incorporate aspects of the cellular environment, high-resolution tracking and manipulation in living cells, and mathematical modeling to understand motor protein dynamics in the cell. In particular, we aim to develop methods to extend the application of single molecule techniques such as optical trapping, FRET, and subpixel resolution tracking to examine motor function in living cells.

Research Description:

Dr R Anne McKinney obtained her Ph.D, at the University of Ulster in 1992 after completing her BSc.(Hons)undergraduate degree in Biomedical Sciences at the University of Ulster, Northern Ireland. She then spent the next 5 years in the Department of Neurophysiology at the Brain Research Institute, University of Zurich as a postdoctoral fellow under the supervision of Profs S.M. Thompson and B.H Gähwiler. In 1998 she obtained her own group at the Brain Research Institute University of Zurich. Dr McKinney has recently joined the Department of Pharmacology and Therapeutics.
Dr. McKinneys principle research interest is the mechanisms involved in development and maintenance of excitatory synapses in the CNS, during physiological and pathological conditions, such as epilepsy and mental retardation. The synaptogenesis and maintenance of synaptic structures, key issues in neuroscience, are still poorly understood despite intensive research efforts. Her group’s studies are concentrated on the hippocampus a brain region thought to be involved in learning and memory. The McKinney lab is using a combination of techniques including, 4-dimensional confocal laser scanning microscopy, analysis of receptor subtype localization using serial electron microscopy, transgenic animals and advanced electrophysiological techniques to investigate the structure and function of dendritic spines and their synapses. These methods allows them assess the structural basis of synaptic function using multiple approaches.

Naguib Mechawar is Associate Professor and Graduate Program Director in the Department of Psychiatry at McGill University, and head of the Neuroanatomy of mood disorders and suicide laboratory at the Douglas Institute. In 2001, he obtained his PhD in Neuroscience at the Université de Montréal, where he studied the development and fine morphometric properties of central neuromodulatory innervations under the supervision of Laurent Descarries. Following postdoctoral fellowships in Jean-Pierre Changeux's laboratory at the Pasteur Institute (2001-2003) and in Rémi Quirion's lab at the Douglas Hospital (2004-2006), he joined the Mcgill Group for Suicide Studies in January 2007 as independent researcher and Assistant Professor of Psychiatry at McGill. At that time, he also became director of the Douglas-Bell Canada Brain Bank. Since 2009, he has also been the director of the Stress, Mood- and Impulsivity-Related Disorders research program at the Douglas Institute.

Dr. Mechawar’s research, which is funded by national and international granting agencies, is mainly focused on the implication of glial cells in depression and suicide. His laboratory studies well-characterized postmortem brain samples with a combination of fine neuroanatomical and molecular approaches. The main objectives of his research are to describe (1) the cellular and molecular features displayed by glial cells in different regions of the human brain; and (2) to determine how these features are altered in major depression and suicide. Current projects include the characterization of astrocytes in the human brain, an investigation of cerebral neuroinflammatory markers in depression, and the examination of white matter composition and organization in depressed suicides having suffered from early life adversity.

Professor Nadig and the PoP lab members study language, social communication and social cognition in the neurotypical population and in people on the autism spectrum. One recent theme of work has been to better understand the enormous variability in language development in autism. Studies in this theme have focused on mechanisms of language learning and impairment and the effects of bilingualism on children with autism.
A second theme of work examines individual differences in the dynamics of social interaction. Here we are developing fine-grained methods to measure dyadic coordination and applying them to study subtle differences that may impact conversation abilities. In intervention settings we are exploring whether coordination with a therapist may be related to treatment outcomes. 
A third theme focuses on developing and evaluating innovative interventions to facilitate social communication and promote the social inclusion of people ASD. Studies have included randomized controlled trials of music therapy for school-aged children with autism, and of a transition program for young adults that emphasizes self-determination.

Research in our lab is focused on the the application of novel MRI methodology in conjunction with biophysical modeling to augment the current understanding of brain anatomy and physiology. In neurological disease, the interplay between sub-voxel, cellular-level components such as microglia and axons executes a critical role in determining disease onset and progression. To investigate these features, our lab utilizes both small animal (7 T) and human MRI/MRS (3 T, 7 T) in conjunction with optical microscopy and advanced image processing methods. In particular, we develop MRI physics acquisition and post-processing techniques for mapping myelin density, axonal packing and axonal geometry.  Equally relevant, our lab also focuses on the advancement of physics, signal processing and hardware methodology for ultra-high field (UHF, > 3T) MRI. We develop computational and hardware/enginnering methods for (a) RF field homogenization (static and dynamic B1+ shimming methods) and (b) B0 shimming (B0 field mapping, gradient Z-shimming and multi-coil shim arrays).

Research in my laboratory focuses in understanding macromolecular structure function relationships, particularly in receptor-ligand interactions. We apply this knowledge to translational research and to experimental therapeutics. We have contributed towards the development of protein mimicry technology (reviewed in Nature Biotechnology, 10: 773; Immunomethods 1: 3-12). Using molecular design, biochemistry, and molecular biology techniques large proteins may be dissected to their smallest functional units. These approaches can be adapted for rational drug design and the development of agonists and antagonists. We developed mimics of antibodies (Science, 253:792), cellular receptors (Proc. Natl. Acad. Sci. (USA), 89:5872-5876), and polypeptide growth factors (antagonists, e.g. J. Biol. Chem., 270:6564-6569; and agonists e.g. Jour. Neuroscience 16: 1308-1316; Chem & Biol. 12:1015-28). The small molecule mimetic ligands are better in vivo at imaging and localizing tumors compared to mAbs (Nature Biotechnology, 14:1120-1122). Structural studies of these small molecule artificial ligands demonstrated that they adopt the predicted configurations (J. Med. Chem. 43: 3541-3548, Jour Biol Chem. 273:23652-23658). Pharmacological studies demonstrated that these small molecule designed agents can be useful as drugs or drug leads. We have succeeded in using these agents in various in vivo models of neurodegeneration (cognitive impairment in ageing, glaucoma) (J. Neurosci, 24:8009-18; J. Neurobiol. 58:341-54) and of cancer (Cancer Res. 2001 61:694-9). In vivo experimental cancer therapeutic studies have shown that our approaches improve the therapeutic index of useful (yet toxic) drugs by many fold, and more importantly help to by pass drug resistance (Oncogene 23:3613-21). Small molecules can be used to identify the relevant sites on receptors that can induce a biological response (Trends in Pharmacological Sciences 21: 93-98. Molec. Pharmacol. 57:385-391. Expert Opinion in Therapeutic Patents 9: 737-751). Using the small molecule ligands we can carry out fine studies of receptor biology (Jour Biol Chem. 275: 9946-9956. Proc. Natl. Acad. Sci. (USA) 96:4067-4072. Jour Biol Chem. 273: 34933-34940. Jour. Neuroscience 17: 6031-6037; J. Biol. Chem 276:1023-1029) such as studies of cross-regulation by different receptor activation sites, and biophysical analyses of ligand-receptor interactions. Recently we extended the concept of ligand mimicry to develop functional ligands of non-protein targets such as gangliosides. This is the first demonstration of a designed, synthetic, functional, small molecule ligand selective for a carbohydrate moiety, and builds on the recent conceptual breakthrough that gangliosides can be true functional cell surface receptors.

We are interested in understanding the molecular bases of mechanotransduction, and the role of mechanosensory neurons in normal and pathological pain transmission. Mechanotransduction, the process through which cells convert a mechanical stimulus into an electrical signal, is of fundamental importance to physiological functions such as our senses of touch (including pain) and hearing, as well as our ability to regulate our hydromineral homeostasis (thirst), baroreflex function and myogenic tone (regulation of blood pressure). Mechanosensitive ion channels are membrane proteins responsible for most mechanotransduction processes, yet their molecular identity has not been fully resolved. Because these channels are involved in several pathologies of the nervous (chronic pain, deafness) and cardiovascular (hypertension) systems, the molecular identification of these channels, and understanding their activation properties may lead to the development of new therapeutic strategies in several clinical areas. The projects in the lab focus on two areas of research: (1) Identification of the molecular mechanisms underlying neuronal mechanotransduction. and (2) Identification of central (spinal cord) neuronal circuits engaged by mechanosensory afferents in control or during chronic pain conditions.

My lab focuses on  deciphering of the genetic basis of childhood neurological disease, with a particular focus on disorders of structural brain abnormalities. Currently, the disorders studied include (1) focal cortical dysplasias (congenital brain malformation responsible for severe epilepsy), (2) schizencephaly (congenital brain malformation associated with variable developmental delay, cerebral palsy and epilepsy), and (3) congenital mirror movements (rare disorder in which affected individuals have involuntary movements on the oppositive side of their body mirroring their voluntary movements). Main technologies used will be whole exome sequencing. Biological validation of candidate genes is done using in-vitro and animal models. I am currently looking for a student to validate a candidate gene for Congenital Mirror Movements using the zebrafish animal model. Understanding the genetic basis of congenital movements allows the unraveling of the mechanisms underlying normal brain development and the crossing of neural fibers across the midline of the nervous system.

The role of the multi-oscillator circadian timing system in physiology and behavior: By employing genetic dissection strategies we want to i) explore the biological significance of peripheral clocks located outside the suprachiasmatic nucleus (SCN) and ii) test the hypothesis that the SCN is functionally compartmentalized. It is believed that circadian clocks in the central nervous system play important roles in the control of various brain functions including the regulation of the sleep-wake cycle, locomotion, body weight, reproduction, and reward processing. We will rigorously examine these links by genetically manipulating clock function in selected brain structures as well as tissues and organs that are targets of neuroendocrine regulation such as the pituitary, gonads and adrenal glands. This research will likely yield fundamental insights in how our internal 24-hour timers effects the nervous system and may thus pave the way for new concepts to tackle mental disorders and other brain dysfunctions
